Tryptophan fermentation and extraction process diagram
Technical Process Description
Fermentation needs syrup from glucose line as main carbon source as well as adding nitrogen source and qualified strain. The reaction will be supported by sterile air through continuous sterilization , via cooling water in coils, controlling certain temperature and air volume.
During production process, in order to maintain the stability of strain performance, it is necessary to separate and purify the production strains regularly, to eliminate the declining strains and select high yield and good strains for production.
By using the metabolic function of strain, the Tryptophan can be accumulated to the maximum extent by means of metabolic control. Through continuous sterilization, air purification, strain culturing, fermentation culturing process, complete the accumulation process of Tryptophan, to achieve high yield of acid, high conversion rate, low residual sugar, low cycle, low energy consumption.
1.2 Extraction
1.2.1 Fermentation solution pretreatment
Tryptophan fermentation is neutral fermentation. In order to prevent the fermentation liquid from being contaminated by bacteria during storage to reduce tryptophan, during the material discharge process, the fermentation liquid is inactivated by the heater, while the fermentation liquid is mixed with dilute sulfuric acid through a static mixer, and the pH value is adjusted to the process value, into the fermentation liquid transfer tank, and then into the extraction workshop for purification process.
1.2.2 Ceramic membrane filtration
The essence of the membrane separation process is the process of separating tryptophan fermentation liquor into tryptophan clear liquor and polymer protein and impurities through ceramic membrane, similar to the screening process, according to the size of the membrane pore size to achieve the purpose of material separation. Materials with molecular diameters greater than 0.05um and less than 0.05um are separated by high pressure generated by high pressure pumps. In the process of tryptophan production, membrane filtration method was used to remove mycelium and impurities greater than 0.05um in the fermentation liquor.
1.2.3 Ion exchange
The clear liquor filtered through the membrane enters the filtrate tank, and the separation system is used to continue purification. During the extraction and production of tryptophan, the functional genes in the resin exchange tryptophan ions in the membrane filtrate, the tryptophan is adsorbed on the resin, and the tryptophan ions on the resin are replaced by NH4+ in the ammonia water.
1.2.4 Deamination
The eluent after separation contains part of ammonia residue. After entering the deammonia tower, the ammonia liquor is separated. After the separation, the ammonia water is recycled and the separated deammonia liquor enters the next process.
1.2.5 Evaporator
When the deamination liquor enters the crystallization evaporator, it is evenly distributed in the shell tube of the evaporator through the distributor. Under the action of gravity, the material is film-like and flows from top to bottom. At the same time, heat exchange occurs between the film of the material and the steam on the outer wall of the pipe, and the water in the material is evaporated, which increases the concentration of tryptophan.
1.2.6 Crystallization
After the material is out, the concentrated liquor containing part of the tryptophan crystal enters the vertical crystallization tank. According to the product characteristics, the crystal is cultured by cooling and isoelectric point method. When the crystal meets the process requirements, then it enters the next process.
1.2.7 Crystal separation
Automatic discharge centrifuge and automatic discharge plate filter press are used for separation and extraction. The separated filter cake enters the drying process and the tryptophan mother liquor returns to the separation process for recycling extraction.
1.2.8 Flash drying
The material enters the dryer, under the strong action of the high-speed rotating agitator, the block material is quickly crushed, and it's fully in contact with the hot air, heated and dried. The dry powder rises with the hot gas, the small particles are discharged from the center of the ring outside the dryer, and the undried or large materials are thrown to the wall by centrifugal force, and fall to the bottom again to be crushed and dried. The dried material is carried by air into the dust collection system for collection and treatment.
