Product Description

Fermentation is the most important method for industrial production of lysine. Its principle is artificially change and control the metabolic pathways of microorganisms to achieve the production of L-lysine, such as controlling the nutrients of culture medium.

Currently, industrial fermentation mainly use various mutant strains of bacteria such as Corynebacterium and Brevibacterium. The main raw materials are starch, molasses, corn and other starchy raw materials .Before entering fermentation tank, these raw materials need to be saccharified into glucose, and the fermentation broth formula needs to be supplemented with biotin.

Lysine fermentation is a typical metabolically controlled fermentation method . By controlling the biotin, L-threonine and L-methionine content, temperature, dissolved oxygen (aeration volume and stirring speed) and other conditions in the culture, L-lysine-producing bacteria can accumulate in large quantities Lysine can convert 70% of sugar raw materials into L-lysine without by-products.

1. Preparation for fermentation:

1.1 S
terile glucose syrup: Since this kind of strains cannot directly utilize starch or dextrin as a carbon source, starch must be hydrolyzed into glucose and continuously sterilized before it can be used for fermentation. In industrial production, this process called saccharification, and the saccharified liquid is starch hydrolyzed sugar. When preparing the culture medium, sterile glucose syrup is added to the fermentation broth as the culture medium base material, or it is fed into the fermentation broth in a continuous fed-flow manner.

1.2 Strain screening and seed preparation: Currently, the mainstream strain is Corynebacterium glutamicum  in this industry. In order to meet large-scale industrial production, a sufficient amount of pure seeds with strong vitality must be available. In lysine fermentation, in order to shorten the adjustment period of fermentation, a relatively high inoculum amount of 5% to 10% is generally used. Therefore, according to the fermentation capacity requirement, it often uses secondary or tertiary seed culture.

1.3 Nutrient broth preparation
: As nutrient broth, organic nitrogen sources and trace elements are filtered and continuously sterilized before they transported to the fermentation tank.

1.4 Air preparation:
Engineering bacteria
 are aerobic microorganisms. They can only grow well and synthesize lysine with sufficient oxygen supply. Before be used for fermentation, the air must pass through a multi-stage filter to ensure the quality of the air, therefore, an oil-free screw air compressor should be used to compress the air.

ntifoamer : It is an additive used in the fermentation industry to eliminate foam, includes natural oils, polyethers, higher alcohols and silicone resins. The most widely used is defoaming agent(polyether) in an amount of about 0.03%. After continuous sterilization, the defoaming agent is added to the fermentation tank intermittently.

Ammonium sulfate solution: Ammonium sulfate enters the fermentation tank after being dissolved and continuously sterilized.
 
2. Fermentation: Industrial fermentation is a process in which microorganisms convert some raw materials nutrients into required products through specific metabolic pathways under appropriate fermentation conditions . Usually, the reaction environment is ensured by adjusting the air inlet, temperature and pH value of the fermentation tank, so that production can proceed smoothly. During this process, sterile glucose syrup, ammonium sulfate solution and antifoamer will also be continuously added to assist production.

3. Fermentation broth acidification: After the fermentation is completed, sulfuric acid is added to the fermentation broth to adjust the acidity of the fermentation broth, which is called 1st Acidity adjustment.
Purpose:
1) An acidic environment can reduce the survival rate of miscellaneous bacteria, prevent lysine in the fermentation broth from reacting with miscellaneous bacteria, and reduce yield.
2)The fermentation broth contains small protein molecules, which will prevent filtration
 and block the filter holes in the next process. But these small proteins will flocculate in acidic environment.
3) 
At the same time, the fermentation broth will be inactivated at high temperatures to eliminate the surviving engineering bacteria. 

4. M
embrane filtration: To extract lysine from mature fermentation broth, it must be filtered or centrifuged to remove bacteria protein molecules, inorganic salts and etc., usually, we use microfiltration membranes for filtration. After the microfiltration, the fermentation broth becomes a cleaner lysine solution. And those filter residues will enter the bacterial protein workshop to make protein feed.


5. Ion exchange (Purification): This system is mainly responsible for purifying lysine solution. It uses a strong acidic cation resin to selectively adsorb lysine cations, and then uses ammonia to elute the lysine adsorbed on the resin. During this process, lysine cations are adsorbed by the resin, and impurities in the lysine solution are removed, thereby improving the purity of lysine. The discharged wastewater is evaporated and crystallized, the wet crystals are dissolved to obtain pure ammonium sulfate solution, which is returned to the fermentation system. The separated ammonium sulfate mother liquor is used to produce compound fertilizer.


6. Deamination: After ion exchange process, the purifying solution contains both low-concentration lysine and ammonium ions , and the concentration of ammonium ions are relatively high. The ammonia needs to be removed from the solution by evaporator, central circulation tube evaporators, membrane evaporators and double-effect evaporators can be used for evaporation, and then the evaporated ammonia gas is collected. After the ammonia gas is separated, it is mixed with the steam condensate to generate ammonia water , which is returned to the ion exchange system for recycling.


7. Concentration: When the deamination liquid enters the crystallization evaporator, it is evenly distributed in the tube of the evaporator through the distributor. Under the action of gravity, the material takes the form of a film and moves from top to bottom. At the same time, heat exchange occurs between the film of the material and the steam on the outer wall of the pipe, and the water in the material evaporates, increasing the concentration of lysine.


8. Acidify and crystallization: Put the lysine hydrochloride concentrate into the neutralization tank, add industrial hydrochloric acid while stirring, and adjust the isoelectric point of lysine . Then put the neutralized liquid into the crystallization tank, pass cold water into the jacket, and slowly cool it. When the temperature of the solution drops to 10°C, crystallize for 10 to 12 hours. When the crystal meets the process requirements, it enters the next process.


9. Separation:
The wet crystals are fed into the centrifuge. Under the strong centrifugal force in the centrifuge chamber, the mother liquor adhering to the surface of the material is thrown out of the chamber . After this process, the lysine crystal particles lose 80% of their moisture.

Drying :
Lysine crystal particles with a moisture content of about 20% are sent into the fluid bed. The material is in contact with dry hot air in the fluid bed, after the moisture in the material reaches the boiling point, then the moisture will convert into gas and discharged from the system, thereby the material is dried. The dried materials are cooled again, and the cooled materials are screened to separate large particles and obtain qualified materials for packaging. The large particles will be crushed and then transport to fluid bed.
